  18. Factors that Affect Enzyme Activity Temperature The optimum temperature occurs when the rate of Contact between the active site and substrate is greatest. In humans this is between 35-40oC Higher temperatures denature the enzyme altering the shape of the active site As temperature increases the rate of contact between the substrate and active site increases
