Myelin Regeneration • Use Oligodendrocytes • (support cells that wrap around axon, secrete myelin) • secretion of Antibody #22 hypothesized to induce remyelation (http://discoverysedge.mayo.edu/de07-4-neuro-lucchinetti/) • cytokines could play a role in destroying myelin and oligodendrocytes (Cytokines, Signal Transduction, and Inflammatory Demyelination: Review and Hypothesis. Ledeen R.W.; Chakraborty G.) • Solution: create a system that will secrete something to neutralize/regulate cytokines OR one that, sensing cytokines, will secrete antibody #22

Faster Firing -Neurons fire via the voltage-induced opening of ion channels, allowing ion flow -reducing voltage required to open ion channels would allow more rapid dispersion of ions - subunits of certain ion channels are known (http://www.iuphar-db.org/DATABASE/VoltageGatedSubunitListForward, PMID 11104501 and 14689447) -Solution: modify voltage-sensitive domains of proteins that affect ion channel conformation to respond to lower voltage

Regrowth of Severed Nerves -When severed neurons decay, they leave behind the neurolemma, a hollow tube which serves as a pathway for regrowing nerves - nerve growth factor is a classified protein that encourages nerve growth (duh) - in the brain, regrowth is inhibited by the rapid formation of scars, the release of factors that prevent remyelation and axon repair, and the lack of growth factor and laminins Solution: engineer cells to bind to the end of the neurolemma and secrete ngfand laminins and absorb factors preventing remyelation and axon repair

  6. Burn Treatment/Prevention • Allow O2 to permeate burnt skin • Would not be viable using bacteria, as burn wounds are extremely susceptible to infection and are therefore thoroughly treated with antibiotics. Modifying skin cells wouldn’t work, as the skin cells in a burn wound are, by definition, dead. • Create biofilm to resist burns a/o mimic healthy skin • Doesn’t seem plausible (layer would be difficult/undesirable to maintain, wouldn’t be fireproof unless it was thick enough to see – eww)
