  1. Today’s Goals • Learn techniques to transfer nematodes by “chunking” • Learn to make a worm “pick” • For moving individual worms • Learn to Pick worms and transfer them from plate to plate

  3. Chunking: • Observe C. elegans under dissecting microscope to make sure they are alive • Sterilize a scalpel • Flame it. No Ethanol. • Cut a chunk of agar and transfer to a fresh, OP50 seeded plate • Make 2 plates per person • Put inverted in incubator in back of class that is NOT on • We will check on the results next class.

  4. Picking Worms • Demo – Make a worm pick • Make 1 per person, Instructor will cut the wire for you. • www.silencinggenomes.org

  5. Practice Picking Worms • Use the technique - • Instructor Demonstration • Video: http://nerdwisdom.com/2007/09/01/the-life-of-the-lab-biologist/ • Pick 5 worms, verify they have survived • Each student in group should pick 5 worms • Show me! • Record in lab notebook

  6. Safety Concerns: • Wear glasses and gloves! • Use great care when flaming worm picks • Only need to flame for a MOMENT • DON’T hold it too long or you will melt the pipet!!! • Be sure to flame pick when finished - or bacterial contamination will result
