  1. Soybean SE Transformation • Induction • Plant and zygotic embryo production (6-8 weeks) • Induction on D40 (6-8 weeks) • Proliferation on solid D20 (4 weeks) • Proliferation on FNL (4 weeks)

  2. Soybean SE Transformation- cont. • II. Gene Transfer, Selection, Proliferation &Maturation • Place 20-25 SE clumps on D20 • Shoot • Continue on D20 (1 week) • Transfer into FNL/H20 for seln. (7–10 weeks) • Isolate green, well formed globular SEs, grow on multi-well plate (5 wks) • Proliferate selected material in FNL (5 weeks) • Mature healthy green embryos matured on SHaM (4-5 weeks) • Desiccation (5-7 days) Select healthy matured • SEs for lipid and/or • other analysis

  3. Soybean SE Transformation- cont. • III. Conversion & Seed Production • Desiccated SEs • Germination (1 week) • Plant hardening (3 weeks) • Transfer hardy plantlets into pots, grow-out in greenhouse • Harvest mature seeds (10-16 weeks)
