Anatomic Pathway • Damaged tissue causes the release of Prostaglandins from Cyclooxygenase Enzymes: • Prostaglandins • Lipid compounds that have a hormone-like effect • Stimulate nerve endings to send pain signals to the brain via the CNS • Cyclooxygenase Enzyme (COX): • COX1 - Maintenance of gastrointestinal tract and blood platelets • COX2 - Related to inflammation and pain responses

  6. Mechanism • NSAIDs inhibit COX enzymes: • Aspirin irreversibly modifies COX enzymes to no longer function • Ibuprofen competitively blocks substrate binding sites on enzymes • AspirinIbuprofen
