  1. EPI DEMIC A Simulated Spread of an Infectious Disease

Today’s Task Today you will work with a partner to perform a lab simulation based on a scenario in which you will explore the simulated spread of an infection This activity will indicate the infected persons and the origin of the infectious disease Make sure you have answered pre-lab questions and turned them in to your instructor

  3. Procedures -Begin by reading the scenario and discussing it with your group. Make sure you understand the problem. Make some predictions about who has the disease -Gather the materials you need or check the materials provided at your station

  4. Procedures -Each station should have: --Permanent Marker --Micro centrifuge or test tube racks which holds 2 tubes per student --1/4 section of full 96 well ELISA tray --1 positive & negative control solution in dropper bottle with a clean pipette for each bottle --Paper towels --Primary Antibody Solution in dropper bottle with clean pipette -- Secondary Antibody Solution in dropper bottle with clean pipette --Washing solution 100ml in washing bottle --Color Reagent Solution

  5. Procedures -Gather the materials you need or check the materials provided at your station Each Group member should have --Safety Glasses --1 empty centrifuge/test tube --1ml simulated body fluid --Access to negative and positive antigen solution --1 plastic transfer pipette

Procedures Lets get started -First Label both of your tubes with your initials -Next place an S on one tube for sharing and Me on the other tube for the tube you will not be sharing -Transfer half of the simulated body fluids to the empty tube with the transfer pipette -The Me tube should be placed in the assigned rack and set aside

Procedures Now it is time to share and this is how you will do it- -Prepare to share your body fluids with 1 person in the class by choosing someone who is not near you -When Instructor gives the signal, mix your body fluids with the chosen person’s fluids in one of the S tubes by using a transfer pipette to remove from one S tube (you or your partner’s) and add to the other

Procedures -Gently mix the body fluids using the pipette by drawing all fluid into the pipette and pushing it back into the tube about 5 times -Use the transfer pipette to remove ½ of the fluid and put it back in the S tube you emptied of you or your partner -Record the name of the student whose fluids you just mixed in space 1 on the ELISA GROUP DATA TABLE provided

Procedures -When Instructor gives the signal repeat the body fluid exchange step with a different partner and record that person’s name on the table as number 2 -When Instructor gives the signal, find a 3rd person to share your body fluids with following the same procedure and record their name on the table in space 3 -Return to your station, place your tube in the rack provided to complete the simulated ELISA

  10. WHO HAS IT???? • Now it’s time to find out how many people are infected • To find our if you are infected, you will become a Biotechnologist and perform an ELISA • (Enzyme-Linked ImmunoSorbent Assay) • Follow directions on the handout

  11. Procedures • Please note: • Each group member acts as a Biotechnologist to perform the ELISA • One assigned member from each group will also act as an Epidemiologist • Report Hepatitis B research and ELISA Data as a team
