  1. Drought Resistance Erich Baker

  2. Most drought stress genes ABA-dependent. • Resource conservation mode • Common to many stress response pathways • Over 300 ABA-induced genes identified (2006), I did ~40

  3. Some Important Drought Stress Pathways to Consider • MYB transcription factors causes increased cuticle wax deposition • CKXs and IPTs regulate growth and water retention • AREB/ABF transcription factors required for successful drought resistance

  4. MYB Transcription Factors • ABA-responsive transcription factor • Binds directly to promoters of genes encoding enzymes for VLCFA synthesis. • VLCFAs deposited to form cuticular wax • Increased wax leads to reduced water loss through transpiration

  5. Col-0 is wt. Myb96-1D is drought resistant mutant. Myb96-1 is drought-susceptible knockout.

  6. MYB in Blueberry • I found MYB orthologsin blueberry • Generated 3 solid sets of SSR primers • MYB only shown to effect leaves and stems, hopefully not berries

  7. Cytokinins • Plant hormones that promote growth • Negative regulators of drought stress pathways • Balance between growth and drought resistance • CKXs and IPTs regulate CK levels

  8. CKXs and IPTs in Blueberry • Found orthologs of both in blueberry • Generated good SSR primers for each • Perhaps ideal mutants could be identified to strike the perfect growth vs. drought resistance balance for a given season

  9. AREBs and ABF • Transcription factors activated by ABA • Upregulated by drought • Activate many genes associated with drought-resistance • Precise function unclear

  10. AREB/ABF in Blueberry • Located all of these genes in blueberry • Generated solid SSR primers for each • Mutants definitely drought vulnerable • Perhaps other mutants could be drought resistant

  11. SSR Primer Results • Located 41 drought stress-related genes on the blueberry genome and generated primers for each • ExtCaM • GapC • HB6 • HOS10 • IPT • MBM1 • MYB96 • P5CS • Pcht28 • PKABA1 • PLC1 • PLPC1 • PUB18 • RD19 • RD21 • RD22 • RD28 • SAM1 • SAM3 • SEH • Sod2 • SS1 • STZ • UBQ1 • ZFHD1 • ERD11 • AREB1 • AREB2 • ABF3 • ANAC019 • ANAC055 • ANAC072 • Apx1 • CDPK1 • CDPK2 • CKX • DREB1D • DREB2A • DREB2B • ERD13 • ERF7
