  3. High Sucrose Soybean Seed PreparationsStep 1 • Measure out one cup of soybeans for each variety called for in the protocol. • Place the soybeans in a beaker and completely cover with water. • Let stand for 18-24 hours.

  14. High Sucrose Soybean TLCActivityStep 3 • After drying, place the plate in a jar containing 91% rubbing alcohol and cap the jar tightly. The amount of alcohol in the jar should not be higher than the 1.5 mm line on the plate.

  15. High Sucrose Soybean TLCActivityStep 4 • Allow the alcohol solvent to rise to the top of the plate. This can take from 1 1/2-2hrs. • Leave in the jar overnight.

  17. High Sucrose Soybean TLCActivityStep 6 • When the plate is completely dry, wear gloves to handle the plates. • Place it in the dipping reagent and quickly remove it. • Use the hair dryer set on high to dry and heat the plate.
