Overview • RNA interference (RNAi) is a system within living cells that helps to control which genes are active and how active they are in which double- stranded RNA (dsRNA) suppresses expression of a target protein by stimulating the specific degradation of the target mRNA. • The RNAi pathway is found in many eukaryotes including animals & is initiated by the enzyme Dicer, which cleaves long double-stranded RNA (dsRNA) molecules into short fragments of ~20 nucleotides. The siRNA will be unwound into two ssRNA, namely the passenger strand and the guide strand. The passenger strand will be degraded, and the guide strand is incorporated into the RNA-induced silencing complex (RISC) which guides the RISC to the target mRNA.
