Swelling Index of all tablets increases as the time proceeds as polymers gradually absorb water due to their hydrophilicity.Increasing concentration of carbopol 934LR leads to more swelling of the tablets. Incorporation of chitosan as a secondary polymer also leads to an increase in swelling index as shown in figure (II). In-vitro drug release study In-vitro release of zolmitriptan from tablets affected by nature and concentration of mucoadhesive polymers. Increasing Carbopol 934LR concentration and incorporation of chitosan lead to more swelling of tablets and increase in thickness and viscosity of gel layer formed at the surface of the tablet which in turn produce longer diffusional path leads to prolonged and more uniform drug release as shown in figure (III). Conclusion The prepared zolmitriptan buccal mucoadhesive tablets adhere to buccal mucous membrane and release the drug in a prolonged and uniform pattern which leads to improvement in patient compliance and drug efficiency . Clarke’s analysis of drugs and poisons, 3rd edition. 2004; 1714. • Introduction • Buccal mucoadhesive tablets are those which are retained in oral cavity and adhere to buccal mucous membrane as shown in figure (I) .They allow drugs to be released and have a direct access to the systemic circulation through the external jugular vein bypassing hepatic first pass metabolism which leads to higher bioavailability (1) . • Zolmitriptan is a serotonin (5HT1) receptor agonist used for the treatment of migraine. It undergoes hepatic first pass metabolism. Absolute oral bioavailability is 40%.Its half life is 2.5 – 3 hr (2) . • Figure (I): Buccal tablet adheres to mucous membrane • Experimental • Zolmitriptan was kindly donated by Nosch Labs Pvt. Ltd., ( India ). • Carbopol 934LR , Chitosan ,PVP K30 ,Magnesium Stearate and Mannitol were purchased from India. • Pilot press tablet compression machine, model CPMD-10 manufactured by Chamunda pharma machinery private limited, Ahmedabad, India. • USP Dissolution Tester, six cups model, Apparatus II, Erweka Apparatebau GmbH, Germany. • Formulae of Zolmitriptan buccal mucoadhesive tablets ( prepared by direct compression ) are shown in table (1). Table (1): The suggested formulae of Zolmitriptan buccal mucoadhesive tablets. • Results and Discussion • In-vitro swelling study • The swelling behaviour of tablets is an important property for uniform and prolonged release of drug and mucoadhesion.It depends on nature and concentration of mucoadhesive polymers .
