  1. FENOBARBITAL Los barbitúricos son depresores del sistema nervioso central, difieren según la dosis, la duración de acción y el margen de seguridadentre la dosis terapéutica y la dosis tóxica

  2. FENOBARBITAL Mecanismo de acción • Potencia los efectos inhibidores del GABA. La dosis requerida para la acción antiepiléptica es menor que la que causa profunda depresión del SNC • Aumenta la conducción del cloro (Cl) • Disminuye la liberación de neurotransmisores calcio dependiente • Disminuye los efectos excitatorios del glutamato

  3. FENOBARBITAL Toxicocinética • Absorción 80% • Comienzo de acción hasta 6 - 10 horas luego de la ingestión • Pico plasmático 6 a 8 horas luego de la ingestión, aunque se puede retrasar

  4. FENOBARBITAL Toxicocinética • Distribución Unión a proteínas plasmáticas 51% Vd: 0.88 L/kg • Metabolismo Es hidroxilado entre un 60 a 75% • Excreción 78 – 87% de una toma única es excretada en orina • Clearence Fenobarbital: 0,093 mL/min/kg

  5. FENOBARBITAL • Vida ½: • Dosis terapéutica 40 a 70 horas en niños • 100 horas en adultos Sobredosis de 4 a 7 días
