This study examines the editing of ErbB2 polyubiquitin chain topology through targeted MRM assays. After immunoprecipitation, ErbB2 proteins were resolved by SDS-PAGE, and specific gel regions were excised and subjected to trypsin digestion. We present diagrams of K48 and K63 polyubiquitin chains bound to ErbB2, highlighting the diglycine signature peptides formed. Furthermore, we analyze relative changes in ErbB2 ubiquitination under different treatments over time, providing insights into its regulatory mechanisms in cellular contexts.
