Dose-Dependent Inhibition of Cytokine Production by Sonicated S. mitis in PBMCs
This study investigates the impact of sonicated S. mitis on cytokine production in peripheral blood mononuclear cells (PBMCs) from six blood donors. PBMCs were stimulated with intact S. mitis at a concentration of 3×10^7/ml, and various doses of sonicated bacteria were added. The cytokines TNF, IL-12, IL-6, IL-8, and IL-10 were measured in the supernatant after 24 hours, while IFN-γ was assessed after 5 days. Results indicate a dose-dependent inhibition of these cytokines, highlighting the immunomodulatory effects of sonicated S. mitis.

PBMC from six blood donors were stimulated with 3 × 107 bacteria/ml of S. mitis CCUG 31661 and graded concentrations of sonicates of the same bacteria were added. Cytokines were measured in the supernatant after 24 h (TNF, IL-12, IL-6, IL-8 and IL-10) or 5 d (IFN-γ).
