Analysis of Rev-erba Protein Levels in 293T Cells with p300 Transfection

This study investigates the effects of p300 on Rev-erba protein levels in transfected 293T cells. Cells were treated with cycloheximide and harvested at specified time points to assess protein levels through Western blot analysis. Both wild-type (WT) and mutant forms of HA-Rev-erba (K591A, K400A) were evaluated in the presence or absence of p300. This allows for the understanding of how p300 influences Rev-erba stability and expression, shedding light on the molecular interactions in cellular contexts.

  1. p300 hs 0 1 2 3 0 1 2 3 Flag-Rev-erba HA-p300 Tubulin Supplemental figure 1: 293T cells were transfected with Flag-Rev-erba in the presence or absence of p300. Cells were treated with cyclohexamide at time 0 and harvested at the indicated tiems. Levels of Rev-erba, p300 and Tubulin were assesed by Western blot with specific antibodies.

  2. WT K591A K400A - + - + - + p300 HA-Rev-erba HA-p300 Supplemental figure 2: 293T cells were transfected with WT HA-Rev-erba or the indicated single mutants in the presence or absence of p300. The levels of Rev-erba and p300 were assesed by Western blot.
