  1. Presentation of Expression of Certain Enzymes in a Panel of Human Cancer Cell Lines By Sara Soth

  2. Background Information Adenocarcinoma Cancer Cell Line P450 Enzymes Antioxidative Enzymes Drug Metabolism (Enzymes) CYP1A1 and CYP1B1 MnSOD Cancer Cell Lines

  3. Background Information Continued P450 Enzymes Includes CYP1A1 and CYP1B1 Catalyze the oxidation of organic substances.

  4. Background Information Continued AntioxidativeEnzymes Lack of enzyme causes oxidative stress, stress will then cause damage or kill cell. Balance needed.

  5. Background Information Continued Drug Metabolism (Enzymes) Equivalent of xenobiotic metabolism. Works with P450 enzymes. Affect on chemotherapy treatment

  6. Background Information Continued MnSOD • Mitochondrial Superoxide Dismutase • mRNA • Protein

  7. Background Information Continued Cancer Cell Lines • A2780 – Ovarian Cancer • Carcinoma • SKOV-3 – Ovarian Cancer • Caucasian Adenocarcinoma • LOVO and DLD-1– Colorectal Cancer • Adenocarcinoma • MCF-7 and MDA-MB-231– Breast Cancer • Caucasian Adenocarcinoma • HeLa – Cervical Cancer • Epitheloid Carcinoma • C33A – Cervical Cancer • Carcinoma

  8. Methodology RNA isolated by Chomczynski and Sacci methods. Grown on cell plates Collected at 60% confluent Put in Tris-glycine gel with SDS-page. Transfer to Nitrocellulose Perform immunodetection with Rabbit polyclonal anti-CYP1B1Ab and MnSODAb Densitometric quantification of band amplification by ImageJ1.46 software for imaging.
