ABSTRACT The Impact of Disintegrant (Ac-Di-Sol) Concentration on Physical Characteristics of Paracetamol Orally Disintegrating Tablet Made By Wet Granulation Method Paracetamol Orally Disintegrating Tablet (ODT) had been developed to fullfill the demand for more patient compliance. Paracetamol ODT more acceptable for patient who may have difficulty in swallowing conventional tablets, especially in pediatric. This dosage form could disintegrate and dissolves rapidly in saliva within a few seconds without the need of drinking water. To produce ODT dosage forms, disintegrant was needed to disintegrate the tablet rapidly. In this study, Ac-Di-Sol as superdisintegrant was added to know its impact on the physical characteristic of paracetamol ODT which was produced by wet granulation method. The concentration of Ac-Di-Sol was 1% (FII), 3% (FIII) and 5% (FIV), with FI as a control formula. The result showed that increasing the concentration of Ac-Di- Sol would increase the hardness and decrease the friability of the tablets. The disintegration time showed an extraordinary profile, where the disintegration time of formula with Ac-Di-Sol 5% was decreased. Statistical analysis showed significant differences of hardness between FIV with FI and FIII, and significant differences of friability between FI with FIII and FIV, FII and FIV. For the disintegration time, there was significant differences between FI and the other formulas. From this study it was concluded that FII with 1% concentration of Ac-Di-Sol was an optimal formula.
