This study conducted RT-PCR analyses to assess CDKC;1 and CYCT1;4 transcripts in mutant plants expressing RNAi constructs. The transcript levels of both genes were evaluated in mutant and transgenic plants using the Arabidopsis UBQ10 gene as a control.

Semiquantitative RT-PCR analysis was performed to determine the transcript levels of CDKC;1 in the cdkc;2-1 mutant and the F1 mutant transfortants harboring a CDKC;1 RNAi construct (cdkc;2-1/CDKC;1 RNAi). The same RT-PCR analysis was performed to determine the transcript levels of CYCT1;4 in the cyct1;5-1 mutant and the F1 mutant transformants harboring a CYCT1;4 RNAi construct (cyct1;5-1/CYCT1;4 RNAi). The ArabidopsisUBQ10 gene was used as a positive internal control.
