Laurea position at the Italian National Cancer Institute, Regina Elena (IRE) Telomere regulation by miRNAs in cancer and ageing Background: Telomeres protect chromosome ends from DNA repair mechanism and are essential for cellular viability and genomic stability. The shortening of telomeres in adult cells provides a molecular mechanism for organismal ageing. Cancer cells are able to escape from “ageing” by the activation of telomerase, a reverse transcriptase that adds back and re-juvinates telomeres of cancer cells. Consequently tinhibiting telomerase or telomere control is a promissing aproach for cancer therapies. DNA-FISH detecting telomeres in human cells YOUR PROJECT: Telomeres are controlled by the balanced action of multiple proteins (shelterin, telomerase,..). You will for the first time identify miRNAs that control telomere function in cancer and aging and study the function of these “telo-miRNAs” in the context of cancer and ageing. Your aim will be to identify “telo- miRNAs with high relevance for future ncRNA based cancer therapies. You will use various methods such as DNA-FISH, miRNA technologies, cloning techniques, Immunocytochemistry, cell culture techniques to accomplish these goals.
