In GD, the undegraded glucosylceramide mainly accumulates in the lysosomes of cells of the reticuloendothelial system, and in particular, macrophages. These hyper activated macrophages develop a characteristic morphology (Gaucher cells). There are different types of Gaucher disease, and signs and symptoms of disease vary widely, even within the same type. Type 1 Gaucher disease is the most common form of this condition. Type 1 is also called non-neuronopathic Gaucher disease because the brain and spinal cord (the central nervous system) are usually not affected. The features of this condition range from mild to severe and may appear anytime from childhood to adulthood. Types 2 and 3 Gaucher disease are known as neuronopathic forms of the disorder because they are characterized by problems that affect the central nervous system. The signs and symptoms of Gaucher Disease include swollen belly due to spleen and liver enlargement, bone pain, easily fractured bones, anemia (low blood counts), fatigue, seizures, swallowing abnormalities, oculomotor apraxia, bleeding and bruising problems, etc. The diagnosis can be confirmed through measurement of glucocerebrosidase activity in peripheral blood leukocytes. A finding of less than 15% of mean normal activity proves to be a diagnostic measure to confirm the presence of Gaucher Disease.
