  1. Abb. 24.8 Überlagerung der Kristallstrukturen der Komplexe von Renin mit den Inhibitoren CGP-38560 24.10(graue Kohlenstoffatome) und Aliskiren 24.12 (hellgrüne Kohlenstoffatome). Mit ihrem peptidähnlichen Grundgerüst binden die Inhibitoren in einer gestreckten, faltblattähnlichen Konformation. 24.10 orientiert seine Benzyl- und Cyclohexylmethylgruppe in die weite S3/S1-Tasche. Für den Entwurf von Aliskiren wurden die beiden hydrophoben Seitenketten von 24.10 miteinander verknüpft. Dafür konnte die Peptidkette aufgeschnitten werden und es entstand ein neuer, polarer N-Terminus.
