  1. Understanding the contribution of traffic related pollution on asthma Biomedical Research Mentoring with Dr. Eric Brandt Reda Baig

  2. Traffic related pollution does not cause asthma but it exacerbates asthma Allergens Diesel Exhaust Particle (DEP) Inflammation results in smaller airways Immune cells Mucus Clogging of the airways ASTHMA Contains anti-inflammatory steroids & bronchodilator

  3. Our goal is to understand how exposure to diesel exhaust particles (DEP) exacerbates house dust mite (HDM) induced allergic responses (asthma attacks) Protocol saline, DEP, HDM or HDM+DEP Day 1 4 6 8 11 13 15 18 20 24H Lung cells Centrifuge on glass slide Assess the different cell types

  4. Exposure to allergen (HDM) = eosinophilic inflammationExposure to DEP = neutrophilic inflammation Saline HDM eosinophils neutrophil HDM+DEP DEP neutrophil

  5. Protocol House dust mite DEP saline, DEP, HDM or HDM+DEP Day 1 4 6 8 11 13 15 18 20 24H Lung tissue RNA cDNA qPCR LUNG EPITHELIAL CELLS chemokines Eotaxin-1 CCR3 RESULT ** Fold increase * BLOOD VESSEL Neutrophils Eosinophils

  6. What I’ve Learned • Writing in a lab notebook • Every detail is important • Lab techniques • Identify and count cells from the airways • Isolate RNA from lung  reverse transcribe into cDNA • Math: Dilutions • Run qPCR and generate graphs of results • Run PCR products on agarose gels • Basic Immunology

  7. Future • Continue with research during the summer and throughout undergrad
