This study investigates the effects of 5-aza-2'-deoxycytidine (5-aza-dC) on the promoter methylation and expression of the CASP8 gene in various breast cancer cell lines, including MCF-7, MB231, and SKBR3. Using methylation-specific PCR (MSP) and bisulfite sequencing, we assessed the methylation status and compared the relative levels of maspin as a potential tumor suppressor. The findings reveal differential responses to 5-aza-dC treatment, contributing to our understanding of epigenetic regulation in breast cancer.
