Co-expression of MCCase Subunits in pCDB3: Impact of pH and Temperature on Enzyme Activity

This study explores the co-expression of the α and β subunits of the MCCase from Pseudomonas aeruginosa, using the plasmid pCDB3 under the T7 promoter. The fusion of the α-subunit (liuD) with a 6-His tag and the β-subunit (liuB) with an S-Tag facilitates protein purification and characterization. We investigate the behavior of the recombinant MCCase enzyme in response to varying pH levels and temperatures, providing insights into its stability and activity, which are crucial for potential applications in biotechnology.

The plasmid contains both subunits of the MCCase from P. aeruginosa under the promoter T7 control. The α-subunit and β-subunit encoded by liuD and liuB genes were fusioned with 6-His and S-Tag epitopes, respectively. B) pH and temperature behavior for the recombinant MCCase enzyme.
