  2. Schizophrenia is • A multifactorial disease • Environment • Genetics • No single gene has been found yet, but recent studies have shown the association with single nucleotide polymorphisms with the disease

ETMOLOGY • Despite the etymology of the term from the Greek roots skhizein (σχίζειν, "to split") and phrēn, phren- (φρήν, φρεν-; "mind"), schizophrenia does not imply a "split mind" and it is not the same as dissociative identity disorder (previously known as multiple personality disorder or split personality), a condition with which it is often confused in public perception. • Coined by eugen bueler

  4. Role of Glutamate • Primary excitatory neurotransmitter in hippocampus and cortex. • Risk of excitotoxicity- accum of glutamate receptors • Excitotoxicity in GRIK3 subunit of Glutamate 7 receptor • T/G SNP --> serine -alanine • If guanine was found this was associated with increased risk of Schizophrenia in Caucasians

  5. Question is: is it the same in the Indian population?

  6. Experiment: • Patients: subtypes:paranoid (70%), undifferentiated (25%), catatonic(5%) • Diagnosis: interview, medical records, clinical observations, history, family history • Lab: DNA extraction.PCR. GRIK3 subunit detected and analyzed.

  7. Results: • GRIK 3 ser310ala polymorphism associated with schizophrenia in Indian patients too • GRIK 3 Alanine allele was found to be increased by 44% compared to their healthy controls • This conclusion GRIK3 association in Indian and western European population with Schizophrenia

  8. Conclusion: • Although in Indians and Europeans Ala310allele plays role in Schizophrenia, • No correlation has been found in Japanese and Chinese population. • Thus the disease susceptibility gene is Geographically Restricted • This again proves that Schizophrenia is a multifactorial disease • Only that some people with Ala310GRIK3 allele have a predisposition to getting Schizophrenia
