This study investigates the protective effects of Med and E2 treatments against Ovx-induced loss of CD28 on CD8+ T cells in both bone marrow (BM) and spleen. Flow cytometry was utilized to measure the levels of CD8+CD28+ T cells across different treatment groups: Sham, Ovx, Med, and E2. With a sample size of 10 mice per group, results are expressed as mean ± SEM and show significant preservation of CD28 expression in the treatment groups compared to the Ovx + vehicle group, with p-values indicating substantial differences.

Supplementary Fig. 2A Supplementary Fig. 2B *** *** ** ** *** *** Med or E2 treatment prevented Ovx-induced loss of CD28 on the BM as well as in spleen CD8+ T cells. (A) CD8+CD28+ T cells in BM, (B) CD8+CD28+ T cells in spleen of Sham, Ovx, Med and E2 were measured by flow cytometry as described in Materials and Methods. N = 10 mice/group; data are presented as mean SEM, **P < 0.01, ***P < 0.001 compared with Ovx + vehicle group.
