  2. 摘要 以DNA为模板合成荧光银纳米团簇,并将其用于癌症细胞的体内外检测。体外检测以检测癌细胞中过量表达的miRNA或者其他标志性小分子;体内检测用Aptamer作为识别标记特异性识别癌症细胞并用于细胞成像。

  3. 用巯基修饰的识别序列识别miRNA • 以C3序列(5’-CCCCAAAACCC-3’)为模板,以AgNO3为前驱体,NaBH4为还原剂,合成荧光银纳米团簇。DNA: AgNO3:NaBH4=1:6:6(DNA浓度为20 μM ,反应环境为20 mM,pH7.0,PBS)。激发波长为547 nm,发射波长为~617 nm。 • 方案一:设计与miR21互补的序列5’端巯基修饰的SH-MC22,并且通过巯基与银的共价作用,淬灭银纳米团簇的荧光来识别miRNA • 方案二:设计与MC22部分互补的序列5’端巯基修饰SH-MC10,由于miR21与MC22的互补序列更长,因此可以将MC10取代下来,使得SH-MC10的巯基变成自由巯基,淬灭银纳米团簇的荧光来识别miRNA 表1.实验中用到的序列
