  3. 분석원리 • Evolis Twin Plus 는 검체 Sampling, 희석, 시약분주, 항온, 세척, Plate이동, Reading 등을 해주는 전자동화 기기이다. Evolis Twin Plus는Windows에서 구동이 되며 Assay는 Bio-Rad Assay에서 수정하여 사용 가능하다. • OD값 측정

  4. - 검체: Serum or plasma - 종목당필요량: 10~20ul (+Dead Volume 150ul) - 준비물: 1)D.W: Prime/Rinse/Dispense and Aspirate 2) Wash Solution 3)Tip 4)Diution Plate(필요시)

  5. 검사방법(menual)*Galactimanan* 1. 검체를Rac에 순서대로 정리하고 뚜껑 open후 튜브를 Sample 수만큼 꽂아놓음. 2. 튜브에 Labeling한다.(5단위로 기록) Heating block 을미리 켜놓는다. 3. Sample 300ul에 Treatment (R7) 을 100ul씩 첨가한다. 4. 미리켜둔HeatingBlock이 120도가 되면 6분간 가열한다. (단백질 제거를 위해) 그 후 10분간 원심한다. 여기까지가 검체의 전처리 과정

  6. 5. 원심을 한 검체는상층액50ul를 사용하며, Well에 conjugate50ul를 넣는다. 6. 파라필름으로실링후1시간 15분간 37도에서 Incubation한다. 7. Washing. (기계에 8번으로 맞춰있음) Washing Solution은 D.W940+Buffer 60 8. TMB Solution 200ul씩 분주. 9. RT에서 암실에 30분간 Incubation. 빛차단이 필수이므로 호일로 감싼다. 10. Stop Solution 100ul 첨가 11. 450nm 에서 흡광도 측정
