  2. The Seven Stages of Tail Regeneration • McLean et al. did an excellent job describing the stages of tail regeneration in the leopard gecko. This was a major strength of the paper, ‘A novel amniote model of epimorphic regeneration: the leopard gecko, Eulepharismacularius.’

  3. How it all begins… • The Gecko is one of the most interesting and mysterious creatures known to man. It possesses many cool features to heal itself. The main one is, tail regeneration. Also sometimes referred to as autotomy, which in which in Greece means, self healing. Here are the steps of gecko tail regeneration.

  4. Step 1: The Initial Injury • Exposed tissues. • Spinal cord retracts and is capped by small blood clot. A. The Gecko clots blood very quickly so that it doesn’t die on blood loss. • Integument begins to collapse around the wound site.

  5. Step 2: The Beginnings of Regeneration • Adjacent muscles and connective tissues retracted. • Blastema appears. • First appearance of Osteoclastsand Chondroclasts.

  6. Step 3: • Angiogenesis begins within blastema. • Axons from dorsal root ganglia of original tail begin to grow into blastema.

  7. Step 4: • Regenerating tail is dome-shaped and wider than long

  8. Step 5: Early • Earliest appearance of presumptive cartilaginous skeleton around ependymal tube

  9. Step 5: Late • Keratinizationand the formation of scales within the epidermis. • First expression of Sox9 in cells of regenerating cartilage. Notice different color, and scales.

  10. Step 6: • Tail is a tapering cone. • Differentiated dermis. • Differentiation of cartilage, muscle, and adipose tissue nearing completion.

  11. Step 7: • Tail is longer than wide. But still not the original color. • Pigmentation begins.
