Screening New Detergents for NMR Studies of Membrane Proteins: A Focus on OmpX

DESCRIPTION

In this study, we investigate innovative detergents for NMR spectroscopy aimed at membrane proteins, specifically OmpX. Utilizing a Gilson Liquid Sample Handler with different capillary tube sizes, we performed [15N, 1H]-TROSY NMR on refolded [2H, 15N]-labeled OmpW and concentrated our analysis on the reconstitution of OmpX from inclusion bodies. We describe our sample preparation techniques, detergent concentrations, and the impact of protein concentration on NMR results. Financially supported through JCIMPT and JCSG initiatives by NIH.
