  1. The Interaction of Garlic With Mediators of Inflammation Prof. A. M. Vollmar Dr. V. M. Dirsch H - P Keiß

  2. Overview • Interaction of garlic powders with NF-B • Results • Summary • Interaction of garlic powders with cytokines • Results • Summary • Influence of garlic powders on endothelial NO-production • Assay setup • Results • Influence of garlic powders on apoptosis • Assay setup • Results • Outlook

  3. Effect of garlic on NF-B activity • Neither spanish nor french garlic powders had an influence on TNF-induced NF-B activity • Garlic sulphur constituents don´t seem to interact with NF-B.

  4. Effect of garlic on TNF liberation • Garlic alone induces TNF liberation. • After LPS treatment garlic increases TNF release.

  5. Effect of garlic on Il-1 liberation • Garlic alone shows weak Il-1 liberation. • Garlic increases Il-1 release in low concentrations. • In high concentrations GPE reduce LPS induced IL-1 levels.

  6. Effect of garlic on Il-10 liberation • Garlic alone induces Il-10 liberation. • Garlic is able to increase Il-10 release after LPS stimulation

  7. NO O2 DAF DAF-2T Influence of garlic powders on endothelial NO-production • Endothelial cells were incubated with garlic powder extracts for 20 h. • Supernatants were collected and subsequently incubated with Arginin,Ca2+-Ionophor A23187 and 4,5-diaminofluorescein. • Fluorescence of the supernatants were determined by spectrofluorimetry

  8. Effect of garlic on NO - liberation Garlic extracts on HUVECs 1.40 1.20 1.00 0.80 Spain x-fold control 0.60 France 0.40 0.20 0.00 µg/ml Control 50 500 50 500 2000 2001

  9. Formazan MTT MTT-Assay Cells used: S-Jurkat, Clone J16 Enzyms of the mitochondria membrane metabolise the yellow MTT to blue Formazan > Assay to distinguish between dead and alive cells

  10. Results I • DADS shows cytotoxic effects at 100 µM • AMSO2 and AMSO show weak cytotoxicity

  11. Results II Garlic extracts show very weak cytotoxic effects

  12. Trouble shooting • GPE showed strong bacterial and fungal contaminations • Different solvants had no effect on contaminations • Supplementation of antibiotics to media has cytotoxic effect on Jurkat cells.

  13. Outlook • Influence of garlic on PAI liberation • Interleukin 10 ??????
