This study investigates the behavior of the replisome as it encounters RNA polymerase (RNAP) head-on. Using advanced imaging techniques, Pomerantz et al. reveal how the replisome adapts to the challenge posed by RNAP, potentially influencing replication fidelity and transcription regulation. The findings contribute to our understanding of genomic integrity and cellular responses to transcription-replication conflicts, shedding light on the fundamental processes that occur during DNA replication.
