Validated RP-HPLC Method for the Determination of Nelaribine in Bulk and Tablet Dosage Form

A novel, simple and economic reverse phase high performance liquid chromatography (RP-HPLC) method has been developed for the estimation of Nelaribine in bulk and tablet dosage form with greater precision and accuracy. Separation was achieved on Cosmiscil C18 column (150X4.6mm i.d.,5-u00b5m) in isocratic mode using Triflouro acetic acid PH-3.6 buffer and Acetonitrile in the ratio of 90:10(v/v) as mobile phase, pumped in to the column at flow rate of 1.0 mL min-1and the detection of eluent from the column was carried out using variable wavelength UV detector at 248 nm. The total run time was 15 min and the column was maintained at ambient temperature. The retention time of Nelaribine was 4.003 min. The standard curves were linear over the concentration range of 25-150 -u00b5g/ml with R2 0.999 and the LOD and LOQ values for Nelaribine were 0.04 -u00b5g/ml and 0.12 -u00b5g/ml , respectively. The percentage recovery was found to be 101.76 'u201c 98.72 %, the % RSD was found to be 0.43. The percentage amount of a marketed tablet formulation of Nelaribine was found to be 101.2 %. The method was validated as per ICH guidelines. Validation studies demonstrated that the proposed RP-HPLC method is simple, specific, rapid, reliable and reproducible. Hence the proposed method can be applied for the routine quality control analysis of Nelaribine in bulk and tablet dosage forms. Simultaneous estimation of lisinopril and hydrochlorthiazide in bulk, pharmaceutical dosage forms and in dissolution sam

The aim of the present work was to develop and validate a simple, efficient, economical RP-HPLC method for the simultaneous estimation of Lisinopril (LIS) and Hydrochlorothiazide (HCTZ) in bulk, dosage forms and in dissolution samples of LIS and HCTZ tablets. Chromatography was performed on Inertsil ODS 3 column (250 x 4.6mm, 5µm) with mobile phase containing 10mM Ammonium acetate: Acetonitrile (80:20% v/v) at a flow rate of 1mL/min and eluents were monitored at 220nm. The retention times of LIS and HCTZ were 3.7min and 7.6min respectively and showed a good linearity in the concentration range of 2-10µg/mL for LIS and 5-25ug/mL for HCTZ with a correlation coefficient of 0.998 and 0.996 respectively. The validation characteristics included specificity, linearity, and limit of detection, limit of quantification, precision, robustness and stability. Validation parameters fulfilled regulatory requirements in all cases. The percent recoveries were ranged in between 98-102, (RSD < 2). The developed HPLC method was successfully used for the simultaneous analysis of LIS and HCTZ in bulk, dosage forms and in dissolution samples of marketed LIS-HCTZ combination tablets.

Method development and validation for the estimation of aprepitant in pharmaceutical dosage forms by RP-HPLC

RP-HPLC method has been developed and validated for quantitative determination of Aprepitant in bulk and pharmaceutical dosage forms. All the parameters of the drug met the criteria of ICH guidelines for method validation. The method is very simple, rapid and economic in nature as the peak is well suitable for routine quality control analysis work. Agilent Polaris ODS C18 (150 X 4.6 cm 5µm) column is used. Mobile phase composition consisted of (80:20 v/v) of Methanol and 10mM Sodium acetate (pH adjusted to 3.0 ± 0.1 with glacial ac etic acid) on isocratic mode. The flow rate of the method is 1.0 ml/min. The retention time of the drug is 4.40 minutes. Calibration standards were prepared in the concentration range of 2.53 to 50.57µg/ml. The wavelength of detection is 210nm. The column temperature is maintained at 25 0C. Several modifications in the mobile phase composition were made in order to study the possibilities of changing the selectivity of the chromatographic system. These modifications included the change of the type and ratio of the organic modifier, pH, flow rate, temperature and stability of Aprepiitant also studied in various stress conditions.
