  1. Microscope Microscope Basics Basics

  2. Parts of the Microscope Drag the box to view the correct answer underneath. Diaphragm Base Always carry a microscope with one hand holding the arm and one hand under the base. Ocular lens(Eyepiece) Body Tube Nosepiece Arm Low Power Objective High Power Objective Stage Stage Clips Coarse Adjustment Fine Adjustment Light

  3. Microscope Parts & Their Functions Drag the box to view the correct answer underneath. Controls the amount of light entering the microscope diaphragm Makes large changes in focus Coarse adjustment Makes small changes in focus Fine adjustment Holds the slide in position Stage clips Has the greatest magnification High power objective Has the least magnification Low power objective

  4. Microscope Care • Always carry with 2 hands (arm and base) • Only use lens paper for cleaning • Do not force knobs • Always store covered • Keep objects clear of desk and cords

  5. What’s my power? To calculate the power of magnification, multiply the power of the ocular lens (eyepiece) by the power of the objective lens. What are the powers of magnification for each of the objectives we have on our microscopes? Fill in the table on your worksheet.

  6. Comparing Powers of Magnification We can see better details with higher powers of magnification, but we cannot see as much of the image. Which of these images would be viewed at a higher power of magnification?

  7. Using the Microscope • Lower stage all the way • Rotate nosepiece and click the low-powerobjective lens (red band, shortest) into position over the stage • Place the slide on the microscope • Look into the eyepiece • Use the Coarse Adjustment knob to locate the object • Use the Fine Adjustment to sharpen the image • Use stage clips to hold slide in place

  8. Using the Microscope • Under low power, position the object you’re viewing at the tip of the pointer. • Move the nosepiece to the medium-power objective. • Use the Fine Adjustment knob to focus. • Move the nosepiece to the high-power objective. • Use the Fine Adjustment knob to focus. • DO NOT MOVE THE SLIDE, or you will have to start over with the low-power objective

  9. 3 – Once you have the image in view, rotate the nosepiece to view it under different powers. Draw what you see on your worksheet! Be careful with the largest objective! Sometimes there is not enough room and you will not be able to use it! Let’s give it a try ... 1 – Turn on the microscope and then rotate the nosepiece to click the red-banded objective (low power objective ) into place. 2 – Place a slide on the stage and secure it using the stage clips. Use the coarse adjustment knob (large knob) to get it the image into view and then use the fine adjustment knob (small knob) to make it clearer. 4 – When you are done, turn off the microscope, put away the slides you used, and put the cover on the microscope.

  10. How to make a wet-mount slide … 5 - Slowly lower the cover slip on top of the drop. Cover Slip Lower slowly 6 – Tap out the air bubbles. 7 - Use a paper towel to blot up any excess water. 1 – Get a clean slide and coverslip from your teacher. 2 - Place the object you are viewing in the center of the slide 3 – Place ONE drop of water on the object. Don’t use too much or the water will run off the edge and make a mess! 4 – Place the edge of the cover slip on one side of the water drop.
