4. Each biological stage/tissue type etc. will yield 3 independent biological samples with independent biological sample #s. Example (hypothetical): embryos, 0-2 hr post-hatch. first sample ----> BS #898 embryos, 0-2 hr post-hatch. 2nd sample ----> BS #899 embryos, 0-2 hr post-hatch. 3rd sample -----> BS #900 • RNAs may be prepared from each of these in several ways. Each RNA prep will have an independent RNA prep ID #. • Embryos synchronized at hatching. We will fix and prepare histograms of stage distribution for each age. • L1 and L2 larvae will be collected without worrying about age. • We will attempt to synchronize L3 larvae at the L2 -> L3 molt. If that proves unfeasible we will revert to time after hatching. If we do that we will need a histogram of larval stage vs. hours post-hatch. • The L3 times were designed to maximize sample during the second half of L3, after the mid-instar transition. • For adults we agreed that males and females will cohabit. The sexes will be separated when the flies are harvested for the RNA preps. We also agreed that there will be no “washout” – i.e. we’ll leave the flies on the food until collection. • We agreed to defer large-scale poly-A preparation until there have been further about quantities.
