Enhanced secretion of TNF-α, IL-6, and IL-1β by isolated LPMNC from patients with ulcerative colitis and Crohn’s disease Katie Hickey Erica Thielen
Introduction • Ulcerative colitis (UC) and Crohn’s disease (CD) are characterized as inflammatory bowel diseases (IBD) • UC and CD are inflammations of the digestive tract caused by • Secretion of proinflammatory cytokines due to impaired down-regulation. • LPMNC isolated from both non-involved and involved UC and CD mucosal cells
Introduction • IBD produces TNF-α, IL-6 and IL-1β • TNF-α increases adhesion molecules • IL-1β produced by activated macrophages • IL-6 increased production in acute phase reaction, enhances LPMNC • Similarities between all
Goal of study • Want to determine which proinflammatory cytokines may be important in the initiation of inflammation of chronic mucosal immune response in IBD. • Study demonstated LPMNC spontaneously secreted amounts of the specific cytokines • PWM used because had highest response
Methods • Colon biopsies from 67 patients to obtain LPMNC • Isolation of mucosa cells • Culturing LPMNC and measuring cytokines
Methods cont. • Concentration of TNF-α, IL-6 and IL-1β secretion by LPMNC • Characterization of LPMNC • Removing macrophages to obtain purified lymphocytes

Figure 4 IL-1β secreted from LPMNC
Figure 5 Contribution of macrophages to cytokine secretion by LPMNC
Conclusion • Summary • PWM was used to stimulate cytokine secretion from mucosal cells • Activated cells produce large amounts of proinflammatory molecules, which contributes to damage w/in intestine • Macrophage greatest impact on IL-6 • Future Experiments
