Investigating Protein Interactions with Bait Sequences in Transcription Assays
This study examines the interactions of hybrid proteins with specific bait sequences using reporter gene assays. We focus on the interaction of the bait sequence with transcription factors that drive expression of AD.TATA.HIS3 and MEL1.UAS reporter genes. We assess both specific and non-specific protein interactions, utilizing the minimal GAL4 promoter. Selection for positive growth on CM–His–Leu media supplemented with 3-AT and X-GAL allows for the identification of successful interactions. This work contributes to our understanding of protein-DNA interactions and transcriptional regulation.
