This study explores the functional implications of NF-κB p65 variants, particularly D94E and D97E, in cellular signaling. Utilizing pCMV constructs tagged with FLAG and other markers, we analyze the cytosolic localization and interactions of these variants with IκBα and p50/NF-κB1. Our findings suggest that mutations in the p65 subunit influence its regulatory roles in immune responses and inflammation. This research contributes to a deeper understanding of NF-κB signaling mechanisms and their perturbations in various diseases.
