  1. Paper review 中山醫學大學 公衛系 陳虹文

  2. Topic • Associations of Candidate Loci ACE,AGT system • Severe H/T • CHD & MI • Associations of genetic polymorphism in lipoprotein • APOC3 → Lipid levels,CHD risk • CYP7 → LDL-C levels

  3. Papers search Pubmed Journal of Lipid Research ELSEVIER Web • Keyword:genotype, genetic polymorphlsm, hypertension, ACE, lipoprotein

  4. Papers A. Association of Candidate Loci Angiotensinogen and Angiotensin-Converting Enzyme with Severe Hypertension: The NHLBI Family Heart Study B. Association of Candidate Loci Angiotensinogen and Angiotensin-Converting Enzyme with Coronary Heart Disease and Myocardial Infarction: The NHLBI Family Heart Study

  5. C. Evidence for Association and Genetic Linkage of the Angiotensin-Converting Enzyme Locus With Hypertension and Blood Pressure in Men but not Women in the Framingham Heart Study D. Association of the Sst-I polymorphism at the APOC3 gene locus with variations in lipid levels , lipoprotein subclass profiles and coronary heart disease risk : the Framingham offspring study

  6. E. Association of the A-204C polymorphism in the cholesterol 7α-hydroxylase gene with variations in plasma low density lipoprotein cholesterol levels in the Framingham Offspring Study

  14. Result A: 1.An interaction betweenACE-D & AGT-T was strongly associated with H/T 2. AGT x sex was significant interaction B: 1.the ACE-D and AGT-Tare statistically associated with CHD and MI; 2.there is a synergistic interaction between the two alleles C:1.the associated with H/T and BP level that ACE-DD,or nearly gene (hGH), 2.there is a sex-specific candidate gene in men but not women

  15. Result D:men S2allels was associations with lower HDL-C, smaller LDL particle size &fasting insulin plasma level↑ women TC,LDL-C,apoB plasma levels↑; bothCHD risk not be manifested E:men the CYP7-C was associated with LDL-C, TC/HDL ratio↑ women no relationship but had lower TG levels both atherosclerotic risk remain uncertain

  16. Conclusion • AGT,ACE,or a nearby gene,is a candidate gene for H/T and CHD,MI • Limitations: not sufficiently controlled for confounding factors • Because stroke,MI,renal failure are among the most serious sequelae of H/T,to clarify the uncertainty regarding the possible association of ACE & AGT with these cardiovascular disease

  17. Conclusion • APOC3-S2 alleles is gender specific associations with lipid, lipoprotein levels • CYP7 A-204C is associated with significant variation in LDL-C Different dietary habits across populations may influence the genotype/lipid association Open interesting insights in the complex interactions between diet, lipid value.insulin resistance and cardiovascular risk
