Evaluation of Osteoclast-T Cell Co-Culture Effects on Bone Marrow Cell Function

This study investigates the interaction between osteoclasts and T cells using bone marrow cells isolated from 4-6 week old mice. After overnight culturing in osteoclast cell culture medium, non-adherent bone marrow cells were co-cultured with purified CD4+CD28null T cells in the presence of M-CSF and RANKL. The culture continued for six days, after which conditioned media were collected for TNF-α ELISA analysis. Additionally, the expression level of tartrate-resistant acid phosphatase (TRAP) in the co-culture was measured via q-PCR, demonstrating significant findings (p < 0.001).

A B *** ** *** *** *** *** For osteoclast-T cell co-culture, bone marrow cells (BMCs) were isolated from 4- to 6-week-old mice by flushing the bone marrow from the excised femora with osteoclast cell culture medium containing 10% FCS. After overnight culture in this medium, non-adherent bone marrow cells were added to purified CD4+CD28null T cells in 24-well plates in osteoclast culture medium containing M-CSF and RANKL and culture was continued till day 6. At the end of experiment conditioned media was collected to perform TNF-α ELISA (A). The transcript level of tartarate resistant acid phosphatase (TRAP) in co-culture was determined by q-PCR (B). ***P < 0.001
