  1. iGEM Meeting with Advisors Summer 2010 - Week 4 6.25.2010

  2. Arsenic PCR-ed out ArsR and PAL from e. Coli. Waiting on backbone to begin digestion and ligation. We re-designed primers for ArsB to include a GC clam and will re-PCR ArsB as soon as those primers are in 

  3. Arsenic New Biobrick parts came in:  • ArsR Sensitive promoter • Gas Vesicles with ArsR sensitive promoter • AIDA membrane protein With Plasmid backbones we will: •  Combine Standard Promoter with Gas Vessicles •  Combine Standard Promoter with Efflux system (ArsB) • Combine Standard Promoter with ArsR TF Parts We Have: ArsR, ArsR Promoter, Gas Vessicles, Pal, AIDA Parts We Need: LamB, ArsB

  4. Gold PCR-ed out GolS, GolB working on GesABC, GolT

  5. Decoder We cloned out OmpA, OmpF, MicA, MicF, and Hfq for the decoder. Also waiting on backbone to start digestion and ligation.   Next week: using riboswitch lock and key to compare to our decoder.  MicF (lane 2) MicA (lane 3)  

  6. Lab Work • We've made 7 500 mL broths of competent cells. This made 90 tubes of competent cells! • The backbone should be ready by the end of today. • We have received more biobrick parts that we are going to streak out soon- most likely this weekend. 

  7. Fundraising • Volleyball- We could host a tournament sometime during this summer and have teams pay for entry. We were thinking of offering a cash prize or gift card to the winner.  • Sweet Corn Festival- we want to have a booth there and sell assorted foods such as dip-n-dots, fried oreos, and maybe some grilled stuff. • Psych surveys- each team member can sign up for 10 hours of paid psych surveys.  • Bake sale- during the school year we could make things like puppy chow and sell it outside of bars during the school year. 

  8. Sponsorship • Requesting Supplies from bio companies including Fisher, Midsci, VWR, etc.
