  1. Mapping Genes and Determining the Allele Frequency of Genotype Through the Loci of Independent Variables Vanessa Grieco Dr. Lin Seminar October 6, 2003

  2. CKR5 and Delta 32 • CKR5 is a chemokine (chemical factor released by immune cell, acts as natural suppressor • CKR5 and receptor are proteins • If Delta 32 base pair deleted then a frame shift of amino acid occurs

  3. 32 magic number Addition or deletion not divisible by three will cause a frame shift. Codons are triplet code which consists of three nucleotides 33 has no frame shift

  4. Recessive versus Dominant • Recessive restriction is expressed with 2 copies • Dominate phenotype with only 1 allele • Allele is and alternative form of a gene • Homozygous (same alleles) • Heterozygous (different alleles)

  5. Mapping of CKR genes Dean et.al. 1996, Science, 273:1856-1862

  6. Genetic Markers and HIV-1 Infection Dean et.al. 1996, Science, 273:1856-1862, Fig.2

  7. Results • CKR5 was the only loci that displayed a significant distortion • No difference in allele frequencies of CKR5 between HIV-1-infected and HIV-1 antibody negative individuals • HIV negative individuals have recessive restriction • Heterozygous progresses disease later if it has one allele Delta 32

  8.
