This lab explores how the light reaction of photosynthesis is influenced by the presence or absence of light and the boiling of chloroplasts. We will use DPIP as a substitute for NADP+ to monitor the reduction process through color change from blue to colorless, utilizing a spectrophotometer to measure absorption of blue light. Cuvettes will contain various conditions (normal, boiled chloroplasts, and blank) to investigate their effects on photosynthesis under controlled experimental setups.

LAB FOUR Photosynthesis Adjust empty spectrophotometer to 0% Adjust with blank cuvette to 100% Add 3 drops chloroplast to your cuvette and immediately measure transmittance. Measure again at 5, 10, and 15 minutes.
