This protocol outlines the detailed steps for grossing a prostatectomy specimen with whole mount sections. Following a color-coded scheme (right-green, left-blue, base-yellow, black-anterior), remove sutures and staples, and ink exposed parenchyma in red to indicate false margins. Amputate the seminal vesicles and vasa deferentia, then serially section the specimen for abnormalities. Carefully transect the bladder base and apex while noting distinct lesions.   1. Grossing Prostatectomy Specimen with Whole Mount Sections Matt Gabbeart

  2. Measure Weigh Ink (follows same color scheme: right-green, left-blue, base-yellow, black-anterior)

  3. Remove sutures and staples. Ink any exposed parenchyma red (indicates false margin)

  4. Amputate seminal vesicles and vasa deferentia. Serial section for abnormalities.

  5. Transect bladder base (proximal urethral margin) Radially section around urethra

  6. Transect apex (distal urethral margin) Radially section around urethra

  7. Section the remaining gland from base to apex (superior to inferior), 5mm or less in thickness Remove any residual staples or calculi

  8. Lay out all slices in proper order from base to apex (Slice 1, 2, 3, etc.) Make note of any distinct lesions Every other slice will be submitted for histological evaluation

  9. With the remaining slices, core out the central parenchyma and submit the capsule. Place all saved tissue in formalin

  10. Cassettes should be in order according to template Double check to make sure all cassettes have correct accession #, part #
