  1. MANGO PLANT BREEDINGRandom Amplified Polymorphic DNA (RAPD) By : Setya Nugraha (H 0810109) Victor Suram (H 0810118) Yogi Prasetyo A (H 0810126)

  2. TAXONOMY • Kingdom : Plantae • Phylum : Magnoliophyta • Class : Magnoliopsida • Order : Sapindales • Family : Anacardiaceae • Genus : Mangifera • Species : Mangifera indica

  3. Abstract The mango is a fleshy stone fruit which is indigenous to the Indian subcontinent, belonging to the genus Mangifera, consisting of numerous species of tropical fruiting trees in the flowering plant family Anacardiaceae. Mango is the second most frequently cultivated tropical fruit worldwide. Most popular varieties of mango fruits are Tommy Atkins (South Africa), Osteen (Spain), Eden (Israel), and Ngowe (Kenya). Mango, together with pistachio and cashew, belongs to the Anacardiae family.

  4. ECONOMIC VALUE In the country remains a favorite fruit mangoes in the season.Overseas mango is an exotic fruit that many fans and including the expensive imported fruits.

  5. MANGO BREEDING METHODS • Propagation by Seed (generative) 1.Open Crosses Identifield by Random Amplified Polymorphic DNA (RAPD) • Propagation through vegetative 1. Okulation 2. Transplantation

  6. Why RAPD??? The RAPD marker is an efficient tool which permits to obtain information on genetic similarity among mango plants and produces specific DNA fragments for a parentage test of hybrids selected in open crosses

  7. Random Amplified Polymorphic DNA (RAPD) Random Amplified Polymorphic DNA (RAPD) is a molecular detection system based on PCR, a molecular technique to detect DNA diversity based on DNA duplication. RAPD is also a DNA marker that utilizes short oligonucleotide random primers (dekamer) to amplify genomic DNA of the organism.

  8. RAPD kinds • Ligase Chain Reaction A DNA amplification technique based upon the ligation of OLIGONUCLEOTIDE PROBES. The probes are designed to exactly match two adjacent sequences of a specific target • Polymerase Chain Reaction In vitro method for producing large amounts of specific DNA or RNA fragments of defined length and sequence from small amounts of short oligonucleotideflanking • DISC-PCR A technique that labels specific sequences in whole chromosomes by in situ DNA chain elongation or PCR (polymerase chain reaction)

  9. The excessand deficiencyof this method • Excess : We can get excellent planting product than before An efficient hand pollination procedure • deficiency : Needs well-trained people This method more sensitive with PCR reaction change
