Cell Counts by Hemocytometer

Cell Counts by Hemocytometer. 9 large squares – 1mm x 1mm Center square divided into 25 smaller squares - 0.2mm x 0.2mm = 0.04 mm 2 Depth of the counting chamber – 0.1 mm Volume of a small square – 0.04 mm 2 x 0.1 mm = 0.004 mm 3 1 ml = 1 cm 3 = 1000 mm 3 (10 mm = 1 cm).

  1. Cell Counts by Hemocytometer

  2. 9 large squares – 1mm x 1mm • Center square divided into 25 smaller squares - 0.2mm x 0.2mm = 0.04 mm2 • Depth of the counting chamber – 0.1 mm • Volume of a small square – 0.04 mm2 x 0.1 mm = 0.004 mm3 • 1 ml = 1 cm3 = 1000 mm3 (10 mm = 1 cm)

  3. 40x magnification 1 mm

  4. 100x magnification 0.2 mm

  5. 400x magnification

  6. Zoox Density Calculations • Average of 10 counts of 0.004 mm3 = 6 zoox • 0.004 mm3 x 1 ml/1000mm3 = 4 x 10-6 ml • 6 zoox/4 x 10-6 ml = 1.5 x 106 zoox/ml • Total homogenate volume = 5 ml • Total zoox = 5ml x 1.5 x 106 zoox/ml = 7.5 x 106 zoox • Wet weight of polyps = 0.25 g • Zoox density: 7.5 x 106 zoox/0.25 g wet wt. = 3.0 x 107 zoox/g wet wt.
