  1. Flow Cytometry

  2. Flow Cytometry Laser

  3. Forward Scatter Forward Scatter indicates size

  4. Side Scatter Side Scatter indicates complexity

  5. Fluorescence Measurements FITC labeled cell! Fluorescent wavelength detector

  6. Fluorescence Measurements PE labeled cell! Fluorescent wavelength detector

  7. Fluorescence Detection • FL-1: FITC, CFSE, ALEXA • FL-2: PE, PI • FL-3: Texas Red, Red-670 • FL-4: APC, Topro

  8. Size and Complexity Separation This demonstrates gating by size and complexity based on forward scatter and side scatter, respectively.

  9. Negative Control No Stain No Stain This sample is a negative control. It has no conjugated stains and represents the overall background.

  10. No Stain CD4:FITC Single Staining This sample has a single stain of CD4 labelled with FITC stain, giving a green color to those cells expressing CD4.

  11. Single Staining CD8:PE No Stain This sample has a single stain of CD8 labelled with PE stain, giving an orange color to those cells expressing CD8.

  12. Double Staining CD8:PE CD4:FITC This sample has a double stain of CD4:FITC and CD8:PE, giving a green and orange color to those cells expressing both CD4 and CD8.
