Concentration Matters

This study emphasizes the critical importance of DNA concentration for reliable sequencing results using the 3130XL. We require 50 ng of DNA in 10 µL, running 5 µL on the sequencer. Notably, samples at 20 ng show enhanced call reliability, improved signal intensity, and better trace scores compared to lower concentrations. The Contiguous Read Length reflects the longest uninterrupted base stretch with a quality score above a set threshold. The QV20+ value quantifies the bases with quality scores ≥ 20, ensuring a 99% probability of accuracy in base calling.
