Molecular Characterization of S. Typhimurium Strains via Colony PCR Analysis

This study employs colony PCR to molecularly characterize S. Typhimurium strains SL7207 (wild type) and SL7207DthyA. The PCR analyses utilize two distinct primer sets: asd primers for strain SL7207 and strain c8799 (Dasd), focusing on a specific deletion, and thyA primers for both SL7207 and its derivative with disrupted thyA. The expected PCR amplification products are confirmed through size markers, indicating successful confirmation of genetic alterations in the strains. Results are illustrated with numerical product sizes.

Molecular characterization of S. typhimurium strains by colony PCR. a) An asd primer set was used for PCR with strain SL7207 (wt) and strain c8799 (Dasd) that encompasses the deletion. b) A thyA primer set was used for PCR with strain SL7207 (wt) and strain SL7207DthyA that encompasses the disruption. Due to the strategy of disruption the two amplification products observed are expected. Numbers indicate sizes of PCR products in bp. M indicates size marker: FX174 HaeIII.
