  1. LL DD Supplemental Figure 5. The circadian clocks ofrglg1 rglg2 mutant and WT plants have a different speed. Promoter activities of a core clock component, CIRCADIAN CLOCK ASSOCIATED 1 (CCA1) and of a well-characterized circadian marker, COLD AND CIRCADIAN-REGULATED 2 (CCR2), were previously shown to be under circadian control via promoter::luciferase (LUC) reporter system (Doyle et al. 2002; Eriksson et al. 2003). CCA1::LUC and CCR2::LUC markers were introduced into rglg1 rglg2 plants, which were compared to WT plants carrying the same transgenes. Plantlets harboring CCA1 promoter::luciferase, and CCR2 promoter::luciferase fusion constructs were entrained in 12 hour light 12 hour dark photoperiods and then put into constant light (LL), or constant darkness (DD). Period length of the free running endogenous clock of mutant plants is significantly shorter, particularly when measured in darkness. Differences between WT and mutant may be connected to changes in hormone levels, as the integration of hormonal signals to the circadian clock was recently demonstrated (Salomé et al., 2006; Hanano et al., 2006).
