Investigating AML1-ETO Expression and Its Effect in HSPC Using Cre-LoxP System

This study explores the expression of AML1-ETO in hematopoietic stem/progenitor cells (HSPCs) using a pMSCV vector system with IRES.GFP for dual expression. We utilize a Cre-LoxP approach to conditionally control the expression of AML1-ETO, tracking the effects on HSPC behavior. Probing mRNA levels reveals insights into relative expression changes among experimental groups. Various constructs, including Cre-YFP and HA-AML1-ETO, are analyzed to clarify their roles in HSPC differentiation and proliferation. Results contribute to understanding AML1-ETO-driven leukemogenesis.
